In order to evaluate the pollution potential of agricultural crops after the construction of Onsan Industrial Complex (non-ferrous metal refineries), concentrations of hazardous heavy metals were analyzed for crop samples (rice, barley, soybean, vegetables and fruits) grown near the Complex in 1978. Although a slight difference was found among the kinds, parts and growing regions of the crops, no definite tendency was observed. The mean/maximum concentrations of crop samples were 0.23/4.0ppm As, 0.4/1.2 ppm Cd, 4.88/12.7ppm Cu, 0.09/0.4 ppm Hg, 3.86/5.0 ppm Pb and 41.3/105 ppm Zn, which may serve as the natural background data for this region.

Subclinical sacroiliitis detected by abdominopelvic computed tomography in Korean patients with Crohn's disease

Background/Aims: Sacroiliitis is a frequent extraintestinal manifestation of inflammatory bowel diseases (IBDs). This study aimed to assess the prevalence of sacroiliitis using a validated screening tool based on abdominopelvic computed tomography (APCT) in Korean patients with Crohn's disease (CD) and examine potential associations between clinical characteristics and sacroiliitis. Methods: One hundred five patients with CD undergoing APCT for any indication at an IBD clinic were matched 1:1 for age and sex with 105 controls without underlying chronic illnesses. Using a validated APCT screening tool that defines sacroiliitis as either ankylosis or a total erosion score (TES) ≥ 3, all computed tomography scans were assessed by two independent, blinded radiologists. We compared the prevalence of sacroiliitis between CD patients and controls and clinical characteristics between CD patients with and without sacroiliitis. Results: The prevalence of sacroiliitis was significantly higher in CD patients than in controls (13.3% vs. 4.8%, p = 0.030). All subjects with sacroiliitis had a TES ≥ 3, but no ankylosis. The assessment of sacroiliitis in APCT showed excellent interreader reliability (Cohen's kappa = 0.933 for presence of sacroiliitis). Sacroiliitis in CD patients was bilateral and asymptomatic. There were no significant associations between sacroiliitis and any demographic data or clinical characteristics in these patients. Conclusions: The prevalence of APCT-detected sacroiliitis in CD patients was higher than that in controls, but the condition was asymptomatic. The clinical significance of asymptomatic sacroiliitis in Korean CD patients remains unclear.

The alveolar destruction associated with emphysema cannot be repaired by current practices. Mesenchymal stem cells(MSCs) are a promising tool for cell therapies and several researches using syngeneic, allogeneic MSCs have demonstrated the therapeutic potential in a variety of applications both in animal models and in humans. However, applications of MSCs produced conflicting results in solid organ transplantation and GVHD. This study aimed to evaluate the immunogenicity of the xenogeneic human adipose tissue derived-MSCs(AD-MSCs) in animal model of elastase-induced pulmonary emphysema. To generate emphysema model, 6-week-old female mice were intratracheally instilled with elastase. Then elastase injected mice were separated three groups of elastase-treated only group, every week and every second week AD-MSCs-treated group. Every week treated group were injected with 1×105 of AD-MSCs i.v. on week 0,1,2,3,4 and every second week treated group were injected on week 0, 2, 4. Then lymph nodes and spleen were harvested and varying amounts of MSCs were added to proliferate previously stimulated T cell in vivo. T cells were harvested after 3 days, analyzed with a FACS. In the analysis, each of all four groups did not induce markers of T cells. There were no differences in average percentage of the anti-CD3+ and anti-CD28+ T cell population. In addition, it was demonstrated that expression of Vβ T cell receptor in each group indicated no significant differences. Human AD-MSCs have low immunogenicity, therefore they have immunologic safety for use in clinical applications.

Safety Evaluation of LB20304 , a New Quinolone Antibiotic

General pharmacology of LB20304, a quinolone antibiotic, were examined in terms of general behaviour, cardiovascular, and central nervous system. LB20304 at oral dose of 2,000 ㎎/㎏ did not induce significant behavioural changes in mice. In contrast with ciprofloxacin, LB20304 at dose of 20 ㎎/㎏, i.v. did not show any observable effects on the blood pressure in rats. Displacement of [³H]muscimol binding to the rat brain synaptic membranes was measured. LB20304 was shown to be about five times less potent than ciprofloxacin in specific GABA receptor binding. Drug interaction between LB20304 and 4-biphenyl acetic acid, an active metabolite of fenbufen, was assessed in mice by measuring convulsion and/or subsequent death. A single oral pretreatment with 4-BPA at 400 ㎎/㎏ increased the incidence of convulsion and death after oral administration of ciprofloxacin at the doses of 25, 50, and 100 from zero of five to three of five, two of five, and four of five, respectively, whereas LB20304 alone or combination with 4-BPA caused neither convulsions nor death at the doses of 12.5, 25, 50, and 100 ㎎/㎏, respectively. Quinolones-induced epileptogenic activities were assessed by a direct intracerebral injection of test articles. The CD_(50) values (nmole) are as follows; 169.47, 35.36, 105.29, and 88.67 for LB20304, ciprofloxacin, ofloxacin, and lomefloxacin, respectively. From these data, LB 20304 at therapeutic doses seems to be much more safe than any other quinolones tested.

The phase behavior and mixture densities were obtained for binary mixtures of polyethylene-co-20.2 ㏖e% butene) copolymer with supercritical n-pentane(n-pentane-h_(12) and n-pentane-d_(12)). The static type experimental apparatus with a high pressure variable volume cell & linear variable differential transducer(LVDT) was used. The density data were obtained for pure n-pentane-htz and n-pentane-d,z at the temperature range from 30 to 150℃ and the pressure range from 71 to 2,415 bac The pure densities were at the range from 0.530 to 0.750 g/㏄ and at the range from 0.588 to 0.896 g/㏄ for n-pentane-d_(12) and n-pentane-d_(12), respectively. And also, the phase boundary between single phase/liquid-liquid coexistence region were obtained for 5.3 wt% poly(ethylene-co-20.2 ㏖% butene)-n-pentane-h_(12) system in the pressure range from 10 to 80 bar at I10, 130 and 150℃ so thus the equilibrium pressure of phase truttsition between single phase[L]/liquid-liquid[LL] coexistence region decreased as the temperature decreased. Also, the mixture densities were obtained in the pressure range from 45 to 2,415 bar at the same temperatures. The phase transition data between single phase/liquid-liquid coexistence region were obtained for 5.1 wt% poly(ethylene-co-20.2 ㏖% butene)-n-pentane-d_(12) mixture in the pressure of below 105 bar at the temperature of 110, 130 and 150 ℃. Also the mixture densities were obtained in the pressure up to 2,415 bar at the same temperatures. The mixture densities of two systems showed sluggish curve and decreased as the temperature increased.

Objectives: The purpose of this study is to isolate a population of multipotent cells from human amnion and decidua, respectively. Methods: Human placentas (gestational age, 30~42 weeks) were obtained after vaginal or cesarean deliveries. Amnions and deciduas were divided mechanically. The collected cells from the amnion and decidua were cultured. Cultured cells were immunophenotypically characterized. The adipogenic, osteogenic and neurogenic differentiation capacities were tested, and their growth kinetics were analyzed. Results: We successfully isolated MSCs from both the amnion and decidua. The phenotype of MSCs cultured from different fetal and maternal parts of the placenta was comparable. The growth kinetics of MSCs derived from amnions and deciduas were similar. Isolated MSCs were differentiated into various cell lines such as adipogenic, osteogenic, myogenic and neurogenic cells. Conclusions: The human amnion and decidua could be an excellent source of MSC because they are easily obtainable after delivery and showed a higher expansion capacity than that of MSCs from adult bone marrow.

Background/Aims: The bone marrow functions not only as the primary B-lymphocyte-producing organ but also as a secondary lymphoid organ for CD4 and CD8 cell responses and a site of preferential homing and persistence for memory T cells. Bone marrow T (BM-T) cells are distinguished from peripheral blood T cells by surface phenotype, cytokine secretion profile, and immune functions. In this study, we evaluated the alloreactive potential of donor lymphocyte infusion (DLI) using BM-T cells in mixed chimerism compared to that using spleen T (SP-T) cells. Methods: Cells were prepared using established procedures. BM-T cells were obtained as a by-product of T-cell depletion in BM grafting and then cryopreserved for subsequent DLI. We performed DLI using BM-T cells in allogeneic mixed chimera mice on post-BMT day 21. Results: When the same dose of T cells, 5-10×105 (Thy1.2+), fractionated from BM and spleen were administered into mixed chimeras, the BM-T group showed complete chimeric conversion, with self-limited graftversus-host disease (GVHD) and no pathological changes. However, the SP-T group showed persistent mixed chimerism, with pathological signs of GVHD in the liver and intestine. Conclusions: Our results suggest that DLI using BM-T cells, even in small numbers, is more potent at inducing chimeric conversion in mixed chimerism than DLI using SP-T cells. Further study is needed to determine whether cryopreserved BM-T cells are an effective cell source for DLI to consolidate donor-dominant chimerism in clinical practice without concerns about GVHD.
